Custom Monoclonal Antibody Development

Cell Technologies Shared Resource has over 25 years of Monoclonal Antibody production.  Shared Resource has produced primarily mouse monoclonal antibodies to various target proteins using native protein or synthetic peptides for animal injections

Typical Project Flow & Time Line

1. Project flow and timeline depends on the nature of the project and differs from project to project. However, in a typical project with immunogenic protein antigens, we will be able to complete the project in about 4-6 months.
2. PI makes Monoclonal Antibody inquiry made by emailing Anh Le (anh.le@cuanschutz.edu).
3. Meeting with Dr. Anderson and Dr. Le to discuss antigen design, immunizations, screening strategies, costs, and timelines.
4. iLabs Request is submitted and IACUC protocol is completed by PI and approved by IACUC (add protocol attachment).
5. Order animals and start immunizations schedule (add immunization schedule attachment).
6. Immunization Timeline 6-8 weeks.

Antibody Express in Mammalian Expression Systems

1. Antibody expression in Mammalian cell lines (Expi 293) is accomplished by transient transfection of light and heavy chain DNA into Expi 293 cells. 
2. Antibody sequences can be obtained from human or animal b-cells.

Antibody Purification

1. CTSR has the capacity to facilitate large scale growth of hybridomas for antibody expression, volumes range from 100mls-5L.
2. Antibody purification CTSR method includes the use of  Sepharose beads which consist of a mix of protein A and protein G beads that have been engineered for high capacity binding and are made specifically for purification on CTSR ATKA pure system.
